The landscape of targeted therapeutics, until recently dominated by Antibody-Drug Conjugates (ADCs), has progressively advanced towards a new class of molecules: X-Drug Conjugates (XDCs).
XDCs represent an innovative approach designed to expand the range of therapeutic tools and improve precision in drug delivery. As the biopharma industry continues to focus on more effective, safer and personalised treatments, XDCs are emerging as a versatile solution that overcomes some of the limitations of existing platforms (1,2), allowing a wide variety of targeted ligands to be paired with different therapeutic payloads. XDCs offer an adaptable design, making them particularly well suited for addressing complex diseases that require selective tissue penetration, precise control of drug release and reduced off-target toxicity.
X-Drug Conjugates (XDCs) combine a targeting moiety (the "X" component) with a bioactive drug payload via a chemical linker. Unlike traditional ADCs, the targeting moiety in XDCs can be an antibody fragment (F-DC), oligonucleotide (O-DC) (3), polymer (Po-DC) (4), peptide (P-DC) (5), aptamer (Ap-DC) (6) , small molecules (SM-DC) or others (7,8), enabling greater flexibility and more specific tissue or cell targeting (Figure 1).
In many ways, XDCs are at the cutting edge of precision medicine, combining the strengths of biologics and small molecules into a single smart therapeutic strategy (9).
XDCs offer many advantages compared to traditional products, such as:
Despite their promise, XDC development deals with challenges such as manufacturing complexity, optimization of linker stability, and difficulty in ensuring favourable safety profiles. Continued research is focusing on scalable synthesis methods, novel payload classes, and improved targeting strategies.
While each XDC offers unique opportunities, they share one common feature: structural and analytical complexity. A thorough characterization is critical to ensure product quality, safety, and regulatory compliance (10).
To elucidate their complex structure, a combination of several analytical techniques can be employed:
